Abstract

To function properly, the immune system must deal withrandomly generated immune receptors that by chancecan recognize self-antigens. Most solutions to this selftoleranceproblem are cell intrinsic and involve the deletionor inactivation of autoreactive cells. This review willpresent an update of the concepts of an extrinsic form ofdominant tolerance mediated by regulatory T cells.Throughout, new tools to identify and manipulate theseregulatory T cells will be discussed. These basics shouldenable us to unlock their clinical potential to specificallysuppress immune responses in autoimmunity and transplantationand to antagonize regulatory T cells where aboost of immunity is required, e.g. in infectious diseasesand anti-tumor immune responses.
