Structure of Bacillus anthracis AtxA, a dimeric virulence regulator, with one subunit color‐coded and one in gray. AtxA contains two phosphoenolpyruvate:carbohydrate phosphotransferase system‐related domains (PRD1 and PRD2; blue and green) and two helix‐turn‐helix (HTH) containing DNA‐binding domains (red and purple). Bacterial iron homeostasis is often regulated by regulators of the Fur family, including in Bacillus subtilis both Fur (left; modeled on PDB 3MWM) and PerR. Resistance to iron (yellow) overload is conferred primarily by induction of a newly assigned iron efflux P1B‐4‐type ATPase, PfeT (top, represented by a P1B‐type ATPase from L. pneumophila PDB 4BBJ) with a secondary contribution from the Fe sequestration protein MrgA (right; represented by E. coli Dps; PDB 1DPS). DdrO regulates the radiation desiccation response and apoptotic‐like cell death in the Deinococcus radiodurans. Light microscopy was used to visualize the effects of depleting the DdrO repressor on D. radiodurans morphology (left to right): Nomarski interference contrast (DIC), membrane staining (FM4‐64), DNA staining (DAPI), and merged images. The cells were depleted of DrdO from 8h (top), 16 h (middle), or 24 h (bottom). DdrO‐depleted cells display morphological changes evocative of an apoptotic‐like response including DNA condensation and vesicle formation. Mutational studies of the xanthine permease XanQ of Escherichia coli, a paradigm for the nucleobase‐cation symporter‐2 (NCS2) family of transporters, identified a critical role for six Gly residues in the gate domain (orange ribbons) and a hydrogen bonding network (inset) involving Gln‐75 (TM2), Asp‐304 (TM9), Thr‐321 (β10) and Gln‐90 (β3) in the core domain.
